NT/2000/XP/Server 2003/Vista/Server 2008 itself will run without it. Really old DOS and other 16-bit apps obviously won't. No 32-bit program will ever make partial use of any of the NTVDM (16-bit) files, so no need to worry about breaking any 32-bit app.
